Everything about Humanin

Description

Humanin is a 24-amino-acid peptide encoded by mitochondrial DNA (16S rRNA gene). The substance was identified in 2001 by research teams around Yuichi Hashimoto as an endogenous anti-apoptotic peptide protecting against Alzheimer-related neurotoxicity.

Per study reports Humanin binds to multiple receptors including the formyl peptide receptor 2 and IGFBP-3. Cellular action includes inhibition of apoptosis, protection against oxidative stress, improvement of insulin sensitivity and neuroprotective effects especially in Alzheimer models.

Humanin levels decline with age - Humanin substitution is being investigated as aging-reverse strategy in preclinical research. In mouse studies extended median lifespan under chronic Humanin application was documented.

Pharmacokinetically limited data - the natural form has very short half-life; synthetic analogs (HNG, Humanin-Glycine-14) were developed for improved pharmacokinetics.

You can order Humanin as lyophilized peptide in 10-pack at 10mg per vial.

Risks & Safety

Humanin is described as well tolerated per study reports. As an endogenous, mitochondrial-encoded peptide, the compound is associated in preclinical studies predominantly with cytoprotective effects.

Observed in research and user reports:

  • Injection site: mild redness or pressure at the injection site is the most common local observation, typical for subcutaneously applied peptides.
  • Very short half-life: the native form has a very short duration of action, which makes pharmacokinetics difficult to control. Synthetic analogs were developed for improvement.
  • Metabolic effects: in animal models, Humanin has been associated with modulation of insulin sensitivity, a systemic metabolic effect beyond pure neuroprotection.
  • No long-term data: the evidence comes predominantly from animal models. Robust long-term human studies do not exist.

This classification does not replace medical advice. Risk and responsibility for any actual use lie entirely with the buyer.

Still have a question?

Write to us, our team will help you.

What is humanin?

A small peptide of 24 amino acids encoded in the mitochondrial genome. It is considered the first of a group of peptides from the mitochondria. It was discovered in 2001 in a search for factors that protect nerve cells from Alzheimer-type damage.

How does humanin work in the cell?

In several ways. At the cell surface it binds to a complex with gp130 and to the receptor FPR2, which beta-amyloid also uses. Inside the cell it binds the protein Bax and prevents it from moving to the mitochondria and triggering cell death.

What do animal studies show on metabolism?

Given into the brain, humanin improved insulin action throughout the body in rats. A stronger variant lowered the blood sugar of diabetic rats by almost 50 percent after one dose. Older mice given the variant HNG had less belly fat and more lean mass.

What is known about the heart?

In an infarction model in mice the variant HNG reduced the size of the infarct depending on the amount. The heart's pumping function was better preserved. This has not been studied in humans.

Does humanin fall with age?

Yes, in mice and humans. In mice the level fell by about 40 percent over the first 18 months of life. People with a particular gene variant in the humanin region had 14 percent less humanin in the blood and showed faster cognitive ageing. Children of centenarians had more humanin than people of the same age.

Does humanin extend life?

In roundworms yes, by just over a day to a mean of 19.0 days. Mice given the variant HNG twice a week from midlife did not live longer. There are no data on this for humans.

Are there human studies?

Not with humanin given as a treatment. ClinicalTrials.gov only lists studies that measure the body's own humanin level. There are therefore no solid data on side effects in humans.
