Description

Kisspeptin-10 is a synthetic decapeptide derived from the KISS1 gene product and characterized in research literature as a central activator of hypothalamic GnRH secretion. The kisspeptin family was identified in the early 2000s as an upstream regulator of the reproductive axis, making the compound class one of the most important discoveries in modern reproductive endocrinology.

The research mechanism lies in the activation of the GPR54 receptor (also KISS1R) on hypothalamic GnRH neurons. In preclinical studies, kisspeptin-10 has been associated with potent stimulation of GnRH release, which downstream leads to LH and FSH increases from the anterior pituitary. This makes kisspeptin-10 a central research tool for studies on the HPG axis (hypothalamic-pituitary-gonadal).

A particularity of the kisspeptin axis: it is essential for the initiation of puberty. Mutations in the KISS1R gene lead to hypogonadotropic hypogonadism, documenting the central role in reproductive activation. In research setups, kisspeptin-10 is therefore frequently used in studies on reproductive axis regulation.

Kisspeptin-10 is the shortest biologically active form of the kisspeptin family (full kisspeptin-54 contains the kisspeptin-10 sequence at the C-terminus). The short form enables more efficient synthetic production and better characterization.

This variant is a ready-to-use nasal spray: 10mg of Kisspeptin-10 dissolved in 5 ml saline and 0.4 ml bacteriostatic water (5.4 ml in total), nothing to mix yourself. One spray (0.1 ml) delivers about 185 mcg. A bottle lasts for about 50 sprays. It is made fresh once you place your order and ships as an express item. As a powder for reconstitution, Kisspeptin-10 remains available in vials.

Risks & Safety

Kisspeptin-10 is described as well tolerated per study reports, but through its direct intervention in the reproductive axis it has its own profile of effects to consider.

Observed in research and user reports:

  • Reproductive hormones: kisspeptin-10 is documented in studies as a potent activator of GnRH, LH and FSH secretion. Noticeable shifts in the reproductive hormone axis are the central expected effect.
  • Nasal lining: the spray adds local effects: brief burning or tingling, an urge to sneeze, a runny nose and a bitter taste in the throat when solution runs down the back. With frequent use a dry lining or slight nosebleeds are possible.
  • Circulation: isolated reports of brief warmth or mild dizziness shortly after application.
  • Very short half-life: the short duration of action of kisspeptin-10 makes pharmacokinetics difficult to control in systemic setups.
  • Limited long-term data: robust human studies exist primarily for short-term endocrine endpoints. Long-term data are largely lacking.

This classification does not replace medical advice. Risk and responsibility for any actual use lie entirely with the buyer.

Write to us, our team will help you.

Is kisspeptin-10 approved?

No. ClinicalTrials.gov lists 9 phase 1 and 4 phase 2 trials with kisspeptin-10, most of them to study the hormone axis. Three small basic-science studies with 14 to 30 people are registered as phase 3. That is why ChEMBL lists phase 3, but they are not approval trials.

How does kisspeptin work?

Kisspeptins are the natural ligands of the receptor GPR54, now called KISS1R. It sits on the GnRH nerve cells in the hypothalamus. People with a non-functional receptor do not go through puberty, their hormone axis does not start. Kisspeptin is therefore the signal at the very start of the chain.

What did the studies show in men?

One dose into a vein tripled LH within 30 minutes. The highest amount tested acted more weakly than the next lower one. An infusion over 22.5 hours raised testosterone by about 45 percent, a lower infusion made LH pulses more frequent. Men with type 2 diabetes and low testosterone also responded.

Does it work the same in women?

No. In the early cycle phase LH did not rise at all in one study, shortly before ovulation it did. After menopause the response was strongest, on the combined pill it was largely absent. The body's own sex hormones clearly dampen or amplify the effect.

What happens with longer dosing?

With 5 days of continuous infusion under the skin testosterone stayed raised, but LH and FSH fell back to control level. With 8 hours of dosing and a 16-hour break each day all three hormones stayed raised over 12 days. There are no longer data.

Is kisspeptin-10 stronger than GnRH?

No. In a direct comparison in healthy men a GnRH infusion raised LH and FSH about three times as much as kisspeptin-10. Kisspeptin-10 and the longer kisspeptin-54 acted similarly. Researchers see the advantage of kisspeptin rather in a more natural stimulation of the axis.
