Stabilised Semax variant

N-acetyl-Semax-amidate, ACTH(4-7) analogue, nootropic in a research context

Half-life
Not measured for NA Semax Amidate. Semax reaches the brain within 2 minutes of a nasal dose and is broken down quickly; the acetyl and amide groups are meant to slow that down

Everything about NA Semax Amidate Nasal

Description

N-Acetyl Semax Amidate is a chemically modified variant of the classical Russian nootropic peptide Semax (Met-Glu-His-Phe-Pro-Gly-Pro). The N-terminal acetylation and C-terminal amidation protect the peptide from enzymatic degradation and substantially extend biological half-life vs unmodified Semax.

Per study reports the substance acts like Semax: modulation of BDNF expression in the hippocampus, neuroprotective effects in cerebral ischemia models, modulation of cholinergic and dopaminergic pathways with cognitive-promoting action. Semax was developed at the Russian Institute of Molecular Genetics and is approved in Russia for treatment of stroke rehabilitation and cognitive disorders.

The acetylated and amidated variant brings improved pharmacokinetics with the same pharmacology - exact comparison data between NA variant and parent compound remain limited.

Semax itself is approved in Russia as nasal drops, so the nasal route is the clinically usual one. There are no human data on the acetylated and amidated form itself.

This variant is a ready-to-use nasal spray: 10mg or 30mg of N-Acetyl Semax Amidate dissolved in 5 ml saline and 0.4 ml bacteriostatic water (5.4 ml in total), nothing to mix yourself. One spray (0.1 ml) delivers about 185 mcg, or about 556 mcg in the 30mg variant. A bottle lasts for about 50 sprays. It is made fresh once you place your order and ships as an express item. As a powder for reconstitution, N-Acetyl Semax Amidate remains available in vials.

Risks & Safety

N-Acetyl Semax Amidate is considered very well tolerated per study reports. The safety profile follows the parent compound Semax, which has consistently been described as low in side effects across decades of Russian research.

Observed in research and user reports:

  • Intranasal irritation: with intranasal use, mild burning, dryness or irritation of the nasal mucosa is the most common observation.
  • Stimulation: occasional reports of mild restlessness, head pressure or shifted sleep, particularly with late-day use, as the compound addresses stimulating and cognitive pathways.
  • No data on the modified variant: exact comparison data between NA-Semax and the parent compound remain limited. Robust long-term human data on the acetylated and amidated form do not exist.

This classification does not replace medical advice. Risk and responsibility for any actual use lie entirely with the buyer.

Still have a question?

Write to us, our team will help you.

How does NA-Semax-Amidate differ from Semax?

Semax is the heptapeptide Met-Glu-His-Phe-Pro-Gly-Pro: a piece of the hormone ACTH plus the end Pro-Gly-Pro. NA-Semax-Amidate additionally carries an acetyl group at the start and an amide group at the end. PubMed has no publication on this form (as of 26 Oct 2026).

Does acetylation change anything about Semax?

In an Italian cell study, yes. Acetylated Semax bound copper differently from Semax and no longer protected nerve cells in culture from damage by copper. Semax with a free start did protect them. Zinc uptake into the cells stayed the same. The amide group of the NA form was not studied there.

How does Semax act in the brain?

In rats a single dose raised the growth protein BDNF in the hippocampus to 1.4 times and activation of its receptor TrkB to 1.6 times. The blueprints of both rose to 3 and 2 times. In a stroke model Semax switched on genes for nerve growth factors and their receptors.

Has Semax been studied in humans?

Yes, mainly after strokes in Russia. In 1997, 30 people received Semax on top of standard care, compared with 80 without Semax. Movement deficits regressed faster. In 2018, blood BDNF rose in 110 people and recovery in daily life went better. None of these studies is listed on ClinicalTrials.gov.

What is known in healthy people?

Little. In a brain scan study with 24 healthy people part of the resting network in the frontal brain was larger 5 and 20 minutes after Semax into the nose than on placebo. Whether focus or memory improve with it was not measured.

Which side effects are known?

There are no solid figures. The studies with Semax give no frequencies in their abstracts. There are no human data on the NA form, only the cell study in which the acetylated form lost its protective effect against copper.
