What is PNC 27?

PNC 27 is an experimental peptide that links a section of the tumor suppressor p53 with a cell-penetrating sequence. In cell models it is meant to selectively destroy cancer cells without attacking normal cells.

Honestly and importantly up front: this is pure basic research in cells and animal models. PNC 27 is not a cancer drug, and there are no human studies. Anyone with a cancer diagnosis belongs in medical care, not on an experimental peptide.

How it works

PNC 27 binds HDM-2, a protein unusually abundant in the membranes of many cancer cells but scarce in normal cells. Through this binding it forms pores in the cancer cell's membrane and triggers its breakdown (necrosis). The selectivity rests on normal cells barely carrying this membrane HDM-2.

  • Bacteriostatic water, U-100 insulin syringes, alcohol swabs.

It was injected in the models. Given the missing human data, particular restraint is warranted here.

What the research shows

A study (Sarafraz-Yazdi, PNAS 2010) showed that PNC-27 binds membrane-bound HDM-2 and destroys cancer cells via pore formation while sparing normal cells. That is an interesting mechanism in cells and animals but no evidence for safe or effective use in humans. PNC 27 is clearly experimental.

Side effects

  • Solid human data are entirely missing.
  • A membrane-active, cell-destroying peptide is not harmless, maximum caution is warranted here.

Storage

Before mixing the powder is robust, normal room temperature (around 20 to 25 degrees) is fine, ideally kept in the dark. After mixing, keep in the fridge (2 to 8 degrees), do not freeze. It stays good there for at least 8 to 10 weeks with barely any loss of potency, in practice considerably longer. Swirl or shake to mix, both are fine.
