Description

Mazdutide (also IBI362 or LY3305677) is an oxyntomodulin analog acting simultaneously on GLP-1 and glucagon receptors. Structurally, it is a synthetic analog of the naturally occurring hormone oxyntomodulin, which addresses both receptor systems in the human body. In research models, the dual activation is associated with a combination of appetite-regulating effects (via GLP-1) and increased energy expenditure (via glucagon).

The compound originates from a collaboration between Innovent Biologics and Eli Lilly, with Innovent leading development in China. Phase 2 studies observed body weight reductions of up to 15.4 percent after 24 weeks. Phase 3 data from the Chinese market is currently available; the substance was approved there for use in 2025.

Pharmacokinetically, Mazdutide is designed for weekly subcutaneous use in research protocols. The half-life is in the range of approximately seven days, supporting the weekly frequency. Maximum plasma concentration is typically reached after around 72 hours.

Within the GLP-1/glucagon class, Mazdutide is one of the few compounds directly modeled on the natural hormone oxyntomodulin. This makes the compound interesting for comparative research using natural oxyntomodulin as a reference.

You can order Mazdutide in 10-vial packs at 5mg and 10mg per vial. The Peptide Desk is one of the few suppliers in DE/EU carrying Mazdutide.

Risks & Safety

Mazdutide is considered generally well tolerated per study reports, although as a GLP-1/glucagon dual agonist the documented profile is shaped by gastrointestinal effects and is dose-dependent in the Phase 2 data.

Observed in studies and user reports:

  • Gastrointestinal effects: nausea, vomiting, diarrhea and constipation are the most commonly documented observations, particularly at the start and with overly rapid dose escalation. Slow titration reduces the incidence.
  • Appetite and fluids: the reduced appetite can lead to decreased food and fluid intake. The literature describes an elevated risk of dehydration.
  • Heart rate: the literature documents mild increases in heart rate attributed to the glucagon component.
  • Muscle mass and injection site: weight loss involves a portion of muscle mass alongside fat mass. Mild irritation at the injection site is documented.
  • No long-term data: most of the available evidence comes from the Chinese market. Robust data on very long-term use do not yet exist.

This classification does not replace medical advice. Risk and responsibility for any actual use lie entirely with the buyer.

Still have a question?

Write to us, our team will help you.

Is Mazdutide approved?

In China, yes. The first approval came in June 2025 for long-term weight management in obesity or overweight with a related condition, type 2 diabetes followed in September 2025. Mazdutide is not approved in the EU or the USA, ChEMBL lists phase 3 as the highest phase. The information here describes the state of research and is not a recommendation for use.

How does Mazdutide differ from Tirzepatide and Retatrutide?

The receptors. Mazdutide acts on GLP-1 and glucagon, Tirzepatide on GLP-1 and GIP, Retatrutide on all three. Mazdutide is derived from the gut hormone oxyntomodulin, which naturally addresses both receptors. A running trial with a higher dose compares it directly with Tirzepatide, there are no results yet.

What did the phase 3 trials show?

In GLORY-1 weight fell by 11.0 and 14.0 percent on average after 48 weeks, on placebo it rose by 0.3 percent. In GLORY-2 with a higher dose it was 16.7 versus 1.5 percent after 60 weeks. In type 2 diabetes HbA1c fell by 1.57 and 2.15 percentage points after 24 weeks (placebo 0.14). Against dulaglutide both doses were ahead for HbA1c and weight.

How long does Mazdutide stay in the body?

In the first phase 1b study in China the half-life was between 6.3 and 16.8 days. An attached fatty acid makes this possible. That fits one dose a week, which is how all trials give Mazdutide.

How high were the doses in trials?

The highest study dose was 16 mg per week, in a phase 1 study and a phase 2 trial in the USA. There weight fell by 18.1 percent after 32 weeks, but 20 percent stopped because of side effects. All trials raise the dose in steps over weeks. The entry describes the same principle: start low and only increase while there is no noticeable effect. This is not a dosing recommendation.

Which side effects were seen?

Mainly gastrointestinal effects, mostly mild to moderate. In GLORY-2 53.1 percent had vomiting, 46.9 percent nausea and 39.4 percent diarrhoea (placebo 1.3, 3.2 and 6.5 percent). In GLORY-1 only 0.5 to 1.5 percent stopped because of side effects. In an early study the complaints occurred mainly in the first dose levels.

Which trials are still running?

Four of the listed phase 3 trials are not finished yet: on fatty liver versus semaglutide, on sleep apnoea, in adolescents and on high blood pressure. Phase 2 trials are running on heart failure with obesity and on fatty liver with inflammation (MASH).
