Description

N-Acetyl Semax Amidate is a chemically modified variant of the classical Russian nootropic peptide Semax (Met-Glu-His-Phe-Pro-Gly-Pro). The N-terminal acetylation and C-terminal amidation protect the peptide from enzymatic degradation and substantially extend biological half-life vs unmodified Semax.

Per study reports the substance acts like Semax: modulation of BDNF expression in the hippocampus, neuroprotective effects in cerebral ischemia models, modulation of cholinergic and dopaminergic pathways with cognitive-promoting action. Semax was developed at the Russian Institute of Molecular Genetics and is approved in Russia for treatment of stroke rehabilitation and cognitive disorders.

The acetylated and amidated variant brings improved pharmacokinetics with the same pharmacology - exact comparison data between NA variant and parent compound remain limited.

Typical application is intranasal in multiple daily drop doses. The NA variant can also be administered subcutaneously.

Observed in research and user reports:

  • Intranasal irritation: with intranasal use, mild burning, dryness or irritation of the nasal mucosa is the most common observation.
  • Stimulation: occasional reports of mild restlessness, head pressure or shifted sleep, particularly with late-day use, as the compound addresses stimulating and cognitive pathways.
  • Injection site: with subcutaneous use, isolated reports of mild redness or pressure at the injection site.
  • No data on the modified variant: exact comparison data between NA-Semax and the parent compound remain limited, and robust long-term human data on the acetylated and amidated form do not exist.
