Description

SLU-PP-332 is a synthetic pan-agonist of the estrogen-related receptors (ERRα, ERRβ, ERRγ), developed by customers at Saint Louis University. The compound is not a peptide but a small molecule and belongs to the class of so-called exercise mimetics, compounds that activate cellular pathways normally stimulated by physical training.

The research mechanism lies in activation of the ERR family of nuclear receptors. In preclinical studies, these regulate gene expression programs for mitochondrial biogenesis, fatty acid oxidation and oxidative phosphorylation. In animal models, SLU-PP-332 has been associated with enhanced mitochondrial function in skeletal muscle and cardiac tissue without the need for physical activity.

In preclinical mouse studies, SLU-PP-332 showed effects on endurance endpoints, fat oxidation and metabolic flexibility. The compound data is relatively recent (published from 2023 onwards), but mechanism coverage is well characterized.

In research setups, SLU-PP-332 is bioavailable as an oral small molecule, which distinguishes the compound from peptidic research tools.

Risks & Safety

SLU-PP-332 is a young compound whose data has only been published from 2023 onwards. The mechanism via the ERR nuclear receptors is well characterized, but human data is entirely absent.

Documented in the preclinical literature:

  • Broad nuclear receptor intervention: as a pan-ERR agonist, the compound activates ERR-alpha, -beta and -gamma and thereby intervenes in several gene expression programs for metabolism and mitochondrial biogenesis.
  • Metabolic changes: animal models document altered fat oxidation, metabolic flexibility and endurance endpoints. The systemic effects of sustained ERR activation in humans are not characterized.
  • No human data: no clinical studies exist. Statements on tolerability, dosing or side effects in humans cannot be derived from the literature.
  • Young data: all the evidence comes from a few preclinical studies of recent years; independent replication is largely outstanding.
