What is Adamax nasal spray?

Adamax Nasal is the Semax derivative Adamax supplied as a ready-mixed nasal spray. The bottle holds 10mg of Adamax dissolved in 5 ml of isotonic saline plus 0.4 ml of bacteriostatic water, 5.4 ml in total, which works out to 1.85mg per millilitre. The metered pump delivers 0.1 ml per actuation, so each spray carries 0.185mg of Adamax (185mcg). On paper that is 54 sprays; after priming the pump roughly 50 remain. There is nothing to reconstitute, measure or transfer. The spray is prepared fresh after you order and ships as an express item.

Adamax itself is a chemically altered form of Semax, the Russian heptapeptide built on fragment 4-10 of the hormone ACTH. It is described as N-acetyl-Semax carrying an extra adamantane group, a bulky, fat-soluble hydrocarbon cage that medicinal chemists often attach to make molecules more stable and better at entering the brain. No published characterisation of Adamax exists; that description comes from the research community. Adamax is not approved anywhere and has no studies of its own. If you would rather dissolve the powder yourself, Adamax is still available as a vial with its own lexicon entry.

How it works

Since Adamax descends from Semax, every assumption about its mechanism is borrowed from the parent molecule. Semax is an analogue of ACTH fragment 4-10 that lacks the hormonal activity of full-length ACTH. Its best documented effect is on neurotrophic factors: Dolotov and colleagues (Brain Research 2006) gave rats a single intranasal dose of 50mcg per kg and found 1.4 times more BDNF protein, 1.6 times more phosphorylated TrkB receptor and a clear rise in BDNF and TrkB messenger RNA in the hippocampus, together with better performance in a conditioned avoidance task. BDNF is the signal that supports neurons during learning, wiring and protection from damage. Effects of Semax on the dopamine and serotonin systems are also described.

The adamantane group is meant to patch two weaknesses of Semax: fast enzymatic breakdown and limited fat solubility. The concept has a precedent. Li and colleagues (FEBS Letters 2010) fitted a different neurotrophic peptide, P21, with adamantane and, after peripheral dosing in normal adult mice, measured improved learning, better short-term and spatial reference memory and more newly formed neurons in the dentate gyrus. Whether the same modification does the same for Semax has never been published. For Adamax that leaves a borrowed mechanism and an open proof.

Uptake through the nose

The nose is the classic route for peptides like Semax and Adamax because part of the dose can travel from the olfactory lining along the olfactory and trigeminal nerves straight into the brain, skipping the bloodstream and the blood-brain barrier. Lochhead and Thorne (Advanced Drug Delivery Reviews 2012) conclude in their review that this route works for proteins and peptides, but that only a small fraction of the dose reaches the brain and that the amount varies widely with technique, volume and the state of the mucosa. For Semax specifically, Shevchenko and colleagues (Russian Journal of Bioorganic Chemistry 2006) used tritium-labelled peptide in rats and detected it in the brain two minutes after a nasal dose, yet only about 0.1 percent of the dose per gram of tissue arrived there. The peptide was quickly cut into smaller fragments, mostly the tripeptide Pro-Gly-Pro.

Two practical points follow. First, a nasal dose needs more active substance than an injection because only part of it gets through. Second, the effect starts fast while the peptide itself is present only briefly; the downstream changes in neurotrophic factors last longer. Whether the adamantane group really extends the residence time has not been measured for Adamax. One or two sprays per nostril (0.1 to 0.2 ml) is a volume the nasal lining handles well; larger volumes at once tend to run down the throat and get swallowed, where peptides are digested instead of acting.

What the research shows

There are no published studies on Adamax itself, neither in animals nor in humans. Everything that can be said with confidence comes from two directions: Semax research and research on adamantane-modified peptides. For Semax the preclinical record is dense. The hippocampal BDNF and TrkB data (Dolotov 2006) and the kinetics after nasal dosing (Shevchenko 2006) are described above. In humans, Lebedeva and colleagues (Bulletin of Experimental Biology and Medicine 2018) used resting-state functional MRI in 24 healthy volunteers and showed that a nasal dose of 1 percent Semax, compared with placebo, altered the topography of the so-called default mode network as early as 5 and 20 minutes later, with a larger anterior component in the medial frontal cortex. That demonstrates that nasal Semax reaches the brain quickly and measurably, but it is not evidence of better everyday performance.

The adamantane side rests on the work of Li and colleagues (FEBS Letters 2010), which shows that an adamantane-containing neurotrophic peptide can promote learning, memory and neurogenesis in mice. The peptide there was neither Semax nor Adamax but the hexapeptide P21. Taken together: the mechanism of the parent molecule is well described and the design principle has worked in a different case. Adamax as a molecule has no evidence of its own. Large Western double-blind trials of nootropic use in healthy people are missing for Semax as well. Anyone who values documented effects is better served by the original Semax, also as a nasal spray.

Side effects

Safety data specific to Adamax do not exist. That needs saying plainly. What can be expected is inferred from Semax and from the nasal route: an irritated or dry nasal lining, sneezing, occasionally a mild burning sensation, plus head pressure or restlessness and poorer sleep when used too late in the day. Semax itself is regarded in the literature as well tolerated and serious adverse effects are rarely described, but data on months of continuous use are lacking. The isotonic saline in the spray is neutral for the mucosa. The only preservative is the small share that comes with the 0.4 ml of bacteriostatic water, far less than in a reconstituted vial, so hygiene at the nozzle matters more.

Several situations argue against use: an acute cold or sinus infection (uptake becomes unpredictable and irritation stronger), injured or recently operated nasal mucosa, anyone under 18, pregnancy and breastfeeding. If you take medication acting on dopamine or serotonin, do not try the combination with a Semax derivative without talking to a doctor. Because the data are thin, the usual rule applies with extra weight: start low, increase slowly, stop if irritation persists.

Handling the spray

The spray arrives freshly prepared and belongs in the fridge straight away. Before the first use, pump once or twice into the air until an even mist appears; after that every actuation delivers 0.1 ml. After each use wipe the nozzle with a clean tissue and put the cap back on so nothing dries in and no germs reach the opening. The bottle is personal: do not share it, not even within the household.

Hold the bottle upright when spraying, do not push the nozzle in deep and aim it slightly outward toward the side of the nostril rather than at the septum, where the lining is more sensitive. If a spray clearly misses, do not top it up; the next dose simply proceeds as normal. If the bottle has not been used for a few days, pump once into the air before using it in the nose again. If the solution turns gel-like, lumpy or smells different, stop using it. Slight cloudiness on its own is not a reason.

Storage

Keep the bottle in the fridge at 2 to 8 degrees, upright, capped and protected from light, for example in its outer box. Do not freeze it: freezing can damage peptides and the pump. Chemically, Semax-type peptides are stable in cold solution for months; the limiting factor for the spray is hygiene, because the solution carries only the small share of preservative that comes with the 0.4 ml of bacteriostatic water. At 5 to 10 sprays per day the bottle is empty after five to ten days anyway, long before that becomes an issue.

For a few hours of transport, say to work, a small cooler bag or simply a jacket pocket is fine; avoid a full day of warmth. If the spray has sat at room temperature for some hours it is not spoiled, just chill it again promptly. If you pause for longer, leave the spray in the fridge and pump once into the air before the next use.
