What is Epitalon nasal spray?

Epitalon (also epithalon or AEDG) is a tetrapeptide made of alanine, glutamic acid, aspartic acid and glycine. It was designed around the year 2000 at the St. Petersburg Institute of Bioregulation and Gerontology to mirror the amino acid make-up of epithalamin, an extract of the pineal gland. Longevity research knows it for two observations above all: telomerase activation in human cell cultures and an influence on melatonin production in the pineal gland.

This variant is not a powder but a finished solution: 10mg of epitalon dissolved in 5 ml of isotonic saline plus 0.4 ml of bacteriostatic water, 5.4 ml in total, filled into a spray bottle with a metered pump. Each actuation delivers 0.1 ml, which is about 185mcg (0.19mg) of epitalon. No reconstitution, no syringes, no needles, the peptide is taken up through the nasal mucosa.

The classic cycle logic does not change: epitalon is used for 10 to 20 days and then paused for months rather than taken continuously. If you know the injectable form, the general background sits in the Epitalon entry; this page is about what is different when you spray it.

How it works

The most cited mechanism is telomerase. In telomerase-negative human fibroblasts, epithalon switched the enzyme back on, telomeres grew longer and the cultures kept dividing past the usual limit. In 2025 an independent group in the UK confirmed that normal epithelial and fibroblast cells respond dose-dependently through hTERT and telomerase. The interesting part doubles as a warning: in breast cancer cell lines the telomeres lengthened as well, but through the ALT pathway.

The second strand is the pineal gland. In pinealocyte cultures epitalon raises the enzymes of melatonin synthesis, in old rhesus monkeys night-time melatonin climbed back up and the glucose and insulin response normalised while young animals did not react. In a small human study in women doing night work with impaired melatonin production, sublingual dosing raised excretion of the melatonin metabolite 6-SOMT 1.7-fold and normalised the expression of the clock genes Clock, Csnk1e and Cry2 in blood cells. That is where the link to sleep and circadian rhythm comes from.

For perspective: these are findings from cell culture, animal models and very small trials. Lifespan extension in healthy humans has not been shown. How a tetrapeptide reaches these targets in the body is still an open question.

Uptake through the nasal mucosa

The nasal lining is thin, richly perfused and bypasses the gut, where a peptide would be broken down at once. Small, water-soluble peptides can cross into the bloodstream from there. A fraction travels along the olfactory and trigeminal routes straight into the brain. For epitalon itself there are animal data: in rats under mild stress, the number of IL-2-containing cells in the hypothalamus was reduced 2 hours after nasal application (after intramuscular injection the effect was recorded at the 24-hour mark), in stressed animals the activity marker c-Fos in the pineal gland rose slightly. As little as 30 ng of nasally applied epitalon changed the firing of cortical neurons in anaesthetised rats within minutes.

Bioavailability is clearly lower than with an injection. Part of the solution drains into the throat, part is cleared by the ciliated epithelium and peptidases in the mucosa degrade another share. How much of a spray reaches the circulation has never been measured for epitalon in humans. That is why a milligram-for-milligram comparison with the subcutaneous form makes no sense.

The concentration of 1.85 mg/ml in isotonic saline is gentle on the mucosa. There are no absorption enhancers, so nothing that irritates the nose either. The 0.4 ml of bacteriostatic water brings only a trace of benzyl alcohol, far less than a bacteriostatic-water vial, so the solution is practically unpreserved, which matters for storage.

Handling the spray

Before the first dose, prime the pump once or twice into the air until a fine mist appears. From then on every actuation delivers the same volume. Blow your nose briefly beforehand so the solution lands on mucosa rather than on mucus.

  • Keep your head upright or tilted slightly forward, not back
  • Place the nozzle in the nostril and aim slightly outward toward the side wall, not at the septum
  • Pump once while breathing in gently, do not sniff hard or the solution runs down the throat
  • Breathe calmly for a few seconds afterwards, do not blow your nose right away
  • Wipe the nozzle and put the cap back on

A slightly bitter taste in the throat means some of the dose drained back, which happens with every nasal spray. Keep the bottle to yourself and do not touch the nozzle with your fingers, because the solution is practically unpreserved.

What the research shows

The evidence is broad but thin in quality. Established in cell culture is telomerase activation in human fibroblasts (2003), now independently confirmed (2025). In mice given the peptide for life, mean lifespan did not change, maximum lifespan rose by about 12 percent and chromosome aberrations in bone marrow fell. In old rhesus monkeys melatonin, the cortisol rhythm and glucose tolerance normalised. In humans there is a retinitis pigmentosa trial with injections next to the eye and the sublingual study on melatonin and clock genes, both small and neither replicated independently.

On the nasal route specifically there are only rat experiments. They show that nasally applied epitalon changes markers in the hypothalamus and the pineal gland but say nothing about dose and effect in humans. The 2025 review sums up 25 years of work and names what is missing: data on short- and long-term toxicity, genotoxicity, carcinogenic potential and drug interactions.

To be blunt: almost everything comes from one institute in St. Petersburg. The UK replication of the telomere data is the first substantial step from outside. Nobody has shown lifespan extension in healthy humans.

Side effects

Across the literature epitalon is described as well tolerated. The human studies reported no notable side effects. The spray adds the usual effects of nasal dosing: brief stinging or tingling, an urge to sneeze, a runny nose, a bitter taste in the throat. Because the solution is isotonic and contains no absorption enhancers this is usually mild and settles within a few days. With an irritated or inflamed lining, nosebleeds or a cold, pause instead.

The theoretical point you should know: epitalon activates telomerase. In breast cancer cell lines it lengthened telomeres through the ALT pathway. What that means for an existing tumour has not been studied. The mouse data showed fewer leukaemias and no rise in spontaneous tumours, which is reassuring but no substitute for a safety trial. With a known or past cancer, caution is the sensible default.

Solid long-term human data are missing. The 2025 review lists toxicity, genotoxicity, carcinogenic potential and drug interactions as open questions in so many words. That is exactly why the cycle structure with long breaks is part of the concept and why the TPD line stays in the sub-milligram range.

Storage and shelf life

The spray belongs in the fridge at 2 to 8 degrees, upright, in the dark and with the cap on. Epitalon in solution stays stable there for months. A short transit at room temperature is not a problem. Do not freeze it, the cycle can damage both the pump and the peptide.

The solution is made up in 5 ml of isotonic saline with only 0.4 ml of bacteriostatic water, so the benzyl alcohol is diluted to less than a tenth and barely preserves anything. Clean handling therefore matters more than with a plain bacteriostatic-water vial: do not touch the nozzle, wipe it after each use, do not share the bottle. If the solution turns gel-like, lumpy or smells different, stop using it. Slight cloudiness on its own is not a reason.

In practice: the bottle is mixed fresh after your order, you take it straight into the cycle and at 2 to 3 sprays a day it is used up after 16 to 25 days. That matches the cycle length, so storage rarely becomes an issue.
