What it’s about

The Longevity Stack combines three mechanistically different compounds from modern longevity research. In preclinical models, these pathways are frequently investigated in parallel because they address distinct hallmarks of aging: pineal-mediated chronobiological regulation, mitochondrial function and senescent cell elimination.

The community typically combines these compounds in setups that address multiple fundamental aging mechanisms simultaneously. In contrast to the Anti-Aging Stack with focus on telomere and redox, the priority here lies on cellular senescence and the mitochondrial axis.

The Compounds

Epitalon is a synthetic tetrapeptide from the Khavinson bioregulator school. In preclinical studies, the compound is associated with effects on telomerase activity, pineal gland function and circadian regulation. An established compound of Russian aging research.

MOTS-C is a mitochondrially encoded peptide derived from the 12S rRNA gene. In research models, the compound is associated with AMPK activation, metabolic flexibility and mitochondrial function. Represents the mitochondrial axis of longevity research.

FOXO4-DRI is a synthetic D-retro-inverso peptide with senolytic activity. In preclinical studies, the compound is associated with selective elimination of senescent cells through disruption of the p53-FOXO4 interaction. One of the most prominent senolytic compounds.

Research Context

In preclinical longevity models, these three axes are frequently investigated complementarily. Epitalon addresses chronobiological and telomere regulation, MOTS-C the mitochondrial and metabolic axis, FOXO4-DRI the senolytic elimination of dysfunctional cells. The stack enables study designs that address fundamental aging mechanisms in a multi-layered way.
