Everything about VIP

Description

VIP (Vasoactive Intestinal Peptide) is a 28-amino-acid neuropeptide originally isolated from the intestine but expressed in tissue-spanning distribution throughout the body. Structurally, VIP belongs to the secretin-glucagon family and shows sequence homology to PACAP (Pituitary Adenylate Cyclase-Activating Polypeptide). In research literature, VIP is established as a multi-function peptide with effects on immune modulation, vascular function and neuroendocrine axes.

The research mechanism lies in the activation of VPAC1 and VPAC2 receptors, which are G-protein coupled and trigger intracellular cascades through cAMP signaling. In preclinical studies, VIP has been associated with anti-inflammatory effects, vasodilation, modulation of T-regulatory cells and neurotrophic properties. This multi-modality makes VIP a complex but mechanistically well-characterized research tool.

In animal models, VIP has been extensively characterized in inflammation models, vascular research setups and neurodegenerative studies. A particularity: VIP shows effects on Th1/Th2 balance in preclinical models and can contribute to a Th2 shift in autoimmune research models.

The half-life of native VIP is very short (a few minutes), making systemic research application challenging. In modern setups, stabilized VIP analogs or local application routes are frequently used.

VIP is available in 5mg vials as 10-vial packs.

Risks & Safety

VIP is described as largely well tolerated per study reports, but through its vasoactive action it has its own profile of effects to consider.

Observed in research and user reports:

  • Circulation: VIP is a potent vasodilator. The literature describes flushing, warmth, low blood pressure or dizziness as circulation-related effects.
  • Injection site: mild redness or pressure at the injection site is the most common local observation.
  • Immune modulation: VIP modulates Th1/Th2 balance in preclinical models and can contribute to a Th2 shift, an active intervention in immune regulation.
  • Very short half-life: native VIP has a duration of action of only a few minutes, which makes systemic use difficult to control.
  • No long-term data: the characterization comes predominantly from preclinical models. Robust long-term human data do not exist.

This classification does not replace medical advice. Risk and responsibility for any actual use lie entirely with the buyer.

Still have a question?

Write to us, our team will help you.

Is VIP approved?

Not as a single drug. ChEMBL lists synthetic VIP (aviptadil) at phase 3, the large trials ran in COVID-19. According to a review a combination of VIP and phentolamine is used for erectile dysfunction.

How does VIP work?

VIP binds to the receptors VPAC1 and VPAC2, which it shares with the related PACAP. There it mainly switches on the cAMP pathway. Nerves and immune cells release it. It widens vessels, stimulates glands and dampens inflammatory reactions.

What did the sarcoidosis study show?

20 patients with active sarcoidosis inhaled VIP for 4 weeks. It was safe and well tolerated, lung cells produced less TNF-alpha and the number of regulatory T cells in the lung rose. The study was open and had no placebo group.

Does VIP help in severe COVID-19?

The large TESICO trial with 471 people found no benefit as an infusion, after 90 days 38 versus 36 percent had died. An earlier trial with 196 people missed its main goal. Inhaled, VIP shortened the hospital stay from 10 to 7.8 days in a small study with 80 people.

What does VIP have to do with migraine?

In a study 21 people with migraine received a 2-hour infusion of VIP or placebo. After VIP 71 percent had a migraine attack, after placebo 5 percent. The temporal artery was widened. VIP and its receptors are therefore seen as possible targets for migraine drugs.

Which side effects are known?

Because VIP widens vessels, the regular entry lists a drop in blood pressure, flushing and dizziness. As a long infusion it often triggered attacks in people with migraine. In the large COVID-19 trial serious events by day 5 occurred in 63 versus 56 percent, without a significant difference.
